Toxoplasma gondii multiplies within parasitophorous vacuole that is not recognized by the primary no oxidative defense of host cells, mainly represented by the fusion with acidic organelles. Recent studies have already shown that hydroxyurea arrested the intracellular parasites leading to its destruction. In the present work we investigated the cellular mechanism involved in the destruction of intracellular Toxoplasma gondii. Anticorpos em bovinos (Bos indicus e Bos taurus) e bubalinos (Bubalus bubalis) inoculados com oocistos de Toxoplasma gondii. Estudo comparativo Arq. Bras. Med. Vet. Zootec.

Três animais de cada espécie (Bos indicus, Bos taurus e Bubalus bubalis) foram inoculados, via oral, com 2×10(5) oocistos de Toxoplasma gondii. Seis outros animais, dois de cada espécie, foram mantidos como testemunhas. A resposta de anticorpos avaliada por meio da reação de imunofluorescência indireta iniciou-se a partir do quinto dia pós-inoculação (DPI) nos zebuínos e bubalinos, e no sétimo DPI nos taurinos. Os títulos sorológicos nos taurinos permaneceram elevados até o final do experimento (70º DPI), alcançando níveis máximos (1:16.384) entre o 42º e 49º DPI. The cross-reactive antigens of Neospora caninum and Toxoplasma gondii are important in the exploration to determine the common mechanisms of parasite-host interaction. In this study, a gene encoding N. caninum apical membrane antigen 1 (NcAMA1) was identified by immunoscreening of a N. caninum tachyzoite cDNA expression library with antisera. from mice immunized with recombinant T gondii apical membrane antigen 1 (TgAMA 1). NcAMA1 was encoded by an open reading frame of 1695 bp, which encoded a protein of 564 amino acids. The single-copy NcAMA1 gene was interrupted by seven introns. Nineteen sera and blood samples from wild feline kept in captivity were tested for Toxoplasma gondii antibody and presence of feline immunodeficiency virus (FIV) DNA, respectively. Eighteen (94.7%) of the them were seropositive for toxoplasma. However, the only negative animal, a Leopardus pardalis, was the only FIV positve. These results suggest that the infection by FIV may have compromised its immune system and interfered with antibody production for toxoplasma.

Characterization of a leucine aminopeptidase from Toxoplasma gondii OAK

The M17 family leucine aminopeptidase (LAP) hydrolyze amino acids from the N-terminus of peptides. Many LAPs from parasitic protozoa including Plasmmodium, Trypanosoma and Leishmania, have been intensely investigated because of their crucial roles in parasite biology. In this study, a functional recombinant Toxoplasma gondii LAP (rTgLAP) was expressed in Escherichia coli and its enzymatic activity against synthetic substrates for aminopeptidase, as well as the cellular localization was determined. Our results indicated that TgLAP is a functional aminopeptidase in the cytoplasma of T. gondii.

Characterization of a novel thrombospondin-related protein in Toxoplasma gondii OAK

Toxoplasma gondii is an obligate intracellular protozoan parasite that invades a wide range of host cells. Upon encountering host cells, the parasite releases a large variety of proteins from secretory organelles, such as micronemes, rhoptries and dense granules. The secretion of microneme protein is essential for parasite invasion. We found that a secreted protein with an altered thrombospondin repeat of Toxoplasma gondii (TgSPATR) was a novel microneme protein, which was different from known microneme proteins that carry thrombospondin repeat domains. As alterações clínicas surgidas a partir do 3º dia após inoculação (DAI) foram: hipertermia, taquicardia, taquipnéia, anorexia, prostração, corrimento nasal e lacrimejamento. Eight pregnant mixed breed bitches, serologically negative for Toxoplasma gondii, were divided into three groups. Group I : bitches 01 and 02 (1.0 x 10(7) tachyzoites of Toxoplasma gondii, subcutaneous route); Group II: bitches 03, 04 and 05 (1.5 x 10(4) oocysts, oral route); Group III: bitches 06, 07 and 08 (as control). Comportamento imunológico e antigênico de cinco amostras de Toxoplasma gondii inoculadas em gatos Ciência Rural

A biologia do Toxoplasma gondii demonstra que o gato é o hospedeiro completo, responsável pela disseminação do parasito. Assim, dois gatos domésticos foram imunizados com cada uma das amostras, VPS (humano), LIV-IV e LIV-V (suíno), CPL (caprino) e CN (felino) de T. gondii. Foram utilizados taquizoítas vivos em inóculos endovenosos de 2x10(6) (1° inóculo} e 4x10(7) (2° inóculo - 35 dias após), exceto a amostra VPS, onde 1 gato morreu no 10° dia com sinais clínicos agudos da doença, nas outras amostras, nenhum sinal clínico foi constatado durante os 6 meses de observação. This study describes the characteristics of 31 children with congenital toxoplasmosis children admitted to the University Hospital of Londrina, Southern Brazil, from 2000 to 2010. In total, 23 (85.2%) of the mothers received prenatal care but only four (13.0%) were treated for toxoplasmosis. Birth weight was <2500 g in 37.9% of the infants. During the first month of life, physical examination was normal in 34.5%, and for those with clinical signs and symptoms, the main manifestations were hepatomegaly and/or splenomegaly (62.1%), jaundice (13.8%), and microcephaly (6.9%). Detecção de anticorpos para Toxoplasma gondii em soro de suínos criados e abatidos em frigoríficos da região da grande Porto Alegre-RS, Brasil Ciência Rural

No presente trabalho, objetivou-se contribuir com dados sobre a freqüência de sororeagentes para Toxoplasma gondii em suínos criados e abatidos na Região da Grande Porto Alegre e fornecer subsídios sobre a importância da transmissão deste protozoário, por suínos. Foram coletadas amostras de 240 suínos em frigoríficos da região. A freqüência de anticorpos anti- Toxoplasma gondii, determinada através da técnica de hemaglutinação indireta, foi de 20 % de soros iguais ou superiores a diluição 1:64. Na técnica de imunofluorescência indireta, foram encontrados 33,75% de soro com diluição iguais a 1:16 ou superiores.

Detection of a high prevalence of antibodies against Toxoplasma gondii in cattle in Northern and Midwestern Brazil Rev Salud Anim.

The aim of this study was to determine the prevalence of Toxoplasma gondii in cattle from the Northern and Midwestern regions of Brazil. Serum samples were collected from 1789 animals and tested by indirect enzyme-linked immunosorbent assay (ELISA). The overall prevalence of T. gondii was 83.40% (1492/1789). The prevalence rates of T. gondii-seropositive animals observed in the states of Pará, Tocantins and, Mato Grosso were 87.45%, 87.79% and 73.06%, respectively. The detection of high prevalence rates of T. gondii in cattle deserves special attention because they are the main source of high biological value protein for humans. This finding indicates the need for further studies on the risk that these animals may pose to public health.

The Amazonian manatee (Trichechus inunguis) is an aquatic mammal that inhabits freshwater environments and is endemic to the Amazon Basin. The presence of Toxoplasma gondii antibodies was investigated in 19 manatees in one rescue unit in the northern region of Peru. Antibodies to T. gondii were detected in 12 (63.2 %) of 19 animals by using the modified agglutination test (titer, 1:25), and no association between sex and age of the animals and the presence of T. gondii antibodies was observed (p < 0.05). The results suggest a contamination by T. gondii oocysts in the aquatic environment where these animals live.
